     摘 要:[目的]制备一种具有良好降解性和成骨活性、可注射的自凝固新型骨修复材料。[方法]制备携载rhBMP2的聚乳酸与聚乙醇酸共聚物(PLGA)微球,并将其与 rhBMP2/磷酸钙骨水泥(CPC)复合,制备出rhBMP2/PLGA微球/CPC复合人工骨。探讨了材料的特性,包括形貌、固化时间、抗压强度及反映材料体外降解速度的指标—体外降解液Ca、P浓度变化,测定复合材料rhBMP2的释药速度及体外诱导MSCs细胞成骨分化的能力。[结果]与单纯CPCrhBMP2相比,复合材料的固化时间少量增加,抗压强度下降明显。体外降解速度及体外释药明显提高,释放的rhBMP2具有骨诱导活性。[结论]rhBMP2/PLGA微球/磷酸钙骨水泥新型复合人工骨是具有良好应用前景的骨修复材料。

    Preparation and characterization of novel injectable selfsetting bone substitute containing of rhBMP2 loaded microspheres

    Abstract:[Objective]To develop a novel injectable in situ settting bone graft substitute with higher degradation rate, more release of rhBMP2/CPC.[Method]rhBMP2 loaded PLGA micropheres, fabricated by using a doubleemulsion, solventextraction [(waterinoil) inwater] technique, was incorporated in rhBMP2/CPC during setting. The processing parameters and fundamental properties included microstructure, setting time, compressive strength, and the in vitro degradation of the composite which was indicated by concentration of Ca and P in the degradation liquid were investigated. Meanwhile, as a delivery system of rhBMP2, the release kinetics and the bioactivity of the released rhBMP2 for the composite were determined in vitro. [Result]The diameter of PLGA microsphere was 253μm±64μm, load density of rhBMP2 was 0.52%±0.14%. Compared with that of rhBMP2/CPC, the setting time increased slightly,the compressive strength decreased significantly,but still stronger than that of the composite when microspheres dissolved. The concentrations of Ca and P in the degradation liquid of composites were higher than that of rhBMP2/CPC at all time points. All implants showed a sustaining release for 28 days, the composite released a greater amount of rhBMP2 than CPC delivery system. Alkaline phosphatase (ALP) ,one of the markers of the osteoblast phenotype, was dramatically stimulated by rhBMP2.[Conclusion] The in vitro degradation and rhBMP2 release can be upgraded by intermixing of the biodegradable rhBMP2 loaded PLGA microspheres with CPC during setting, therefore, the combination of rhBMP2 loaded PLGA microspheres and CPC forms a promising synthetic bone graft. ......
