【摘要】 目的 探讨哮喘小鼠骨髓来源树突状细胞(DCs)的表型及共刺激分子在哮喘发病中的影响。方法 用卵清蛋白建立哮喘模型,利用rmGM-CSF和rmIL-4体外诱导骨髓细胞分化为DCs,流式细胞仪检测DCs表面共刺激分子的表达,混合淋巴细胞反应检测DCs刺激同种异体的T淋巴细胞增殖的能力。结果 在两种细胞因子的作用下,从骨髓中可以诱导出大量的DCs;且均表达DCs的表面标志(33D1)。哮喘组DCs表达CD86分子的水平高于对照组,而CD40、CD80在两组之间没有差异;哮喘组与对照组DCs均能强烈刺激同种异体T淋巴细胞的增殖,哮喘组DCs的刺激能力明显强于对照组。结论 哮喘组DCs的抗原递呈能力增强,表明DCs在哮喘的发病中可能起着重要作用。【关键词】 哮喘;树突状细胞;B7;CD40;混合淋巴细胞反应
The study of phenotypes and costimulatory molecules of dendritic cells in asthmatic mice

【Abstract】 Objective The purpose of this study was to evaluate the effects of dendritic cells derived from bone marrow in the pathogenesis of asthma.Methods Murine model of ovalbumin (OVA)-allergic asthma was established.DCs were developed by incubating bone marrow cells with rmIL-4 and granulocyte-macrophage colony-stimulating factor. Phenotype was assessed with flow cytometry, and the antigen-presenting function was assessed with the mixed leukocyte reaction.Results DCs expressed DCs characteristic 33D1 can be developed from bone marrow cells by using rmIL-4 and rmGM-CSF.The stimulating effect of the DCs on the proliferation of the lymphocytes was shown in asthmatic and control group, but the effect in asthmatic group was more intensive than control.DCs from asthmatic mice showed phenotypic differences in the expression of CD86,but there were no differences in the expression of CD80 and CD40 between asthmatic and controlled group.Conclusion This study suggests the antigen-presenting function of DCs in asthmatic group was increased and DCs may play an important role in the pathogenesis of asthma. ......
