睾酮联合绒毛膜促性腺激素建立多囊卵巢大鼠模型的研究

【摘要】 目的 本实验用睾酮联合绒毛膜促性腺激素(HCG)及炔诺孕酮联合HCG两种方法分别建立多囊卵巢大鼠模型,将模型鼠的卵巢形态及血清激素方面改变与人类多囊卵巢综合征(PCOS)的病理改变加以比较,探讨睾酮联合HCG是否是建立多囊卵巢大鼠模型的有效方法,并比较两种造模法是否存在异同点,判断哪一种造模方法更为理想及简便有效。方法 将23日龄SD雌性大鼠随机分为3组,A组皮下注射睾酮及HCG;B组皮下包埋炔诺孕酮避孕棒后注射HCG;C组皮下注射等量生理盐水,20天后称体重,取血,处死取卵巢。用ELISA法检测血激素的改变,称卵巢重量,用光镜观察卵巢形态学改变。结果 A、B组卵巢增大，呈多囊性改变，A、B组与C组比较雄激素增高，LH/FSH增加，但A、B两组之间差异无显著性。结论 睾酮联合HCG是建立多囊卵巢大鼠模型的有效方法，与炔诺孕酮联合HCG的造模法相比更为简便可行。【关键词】 多囊卵巢综合征；大鼠模型；睾酮联合HCG
Study of rat model with polycystic ovary induced by testosterone plus human chorionic gonadotropin

【Abstract】 Objective To establish rat model with polycystic ovary with two different methods:testosterone(T) plus human chorionic gonadotropin (HCG) and progestogen(P) plus HCG，compare the rat models induced by testosterone plus HCG with the patients with polycystic ovary syndrome(PCOS) in the aspects of ovarial morphous and gonadal hormone,and investigate whether testosterone with HCG is the effective method to establish rat model with polycystic ovary syndrome,than compare two methods(T+HCG and P+HCG)finally to draw a conclusion if there are any difference or similary between the two methods and which one is more ideal and convenient.Methods 60 SD female rats of 23 days were divided into three groups in random.Group A:have a hypodermic injection with testosterone plus HCG;Group B:hypodermic embed progestogen contraceptive stick followed by hypodermic injection of HCG;Group C:hypodermic inject same volume of isotonic Na Chloride as control.After 20 days the blood of 60 rats were abstracted by eyeball dissection,gonadal hormone lever were tested with ELISA, then the rats were killed,the rats ovaries were dissected, morphological changes in ovaries were observed by microscopy.Results The ovaries from group A and group B were larger than those from group C and appeared a polycystic change.Testosterone concentration and the ratio of LH/FSH in group A and group B were higher than those of group C,and there was no significant difference between group A and group B.Conclusion Testosterone plus HCG is an effective method to establish polycystic ovary model in rat,furthermore it is more simple compared with the method of progestogen plus HCG. ......
