巨噬细胞集落刺激因子对破骨细胞生物作用的实验研究

     [摘要]目的 探讨在实验条件下巨噬细胞集落刺激因子(M.CSF)在骨代谢过程中对破骨细胞的生物作用。 方法 用不同浓度的M.CSF和活化核因子κB的受体配体(RANKL)与鼠骨髓细胞培养,在加入或不加入牛骨片的情况下诱导产生破骨细胞。分别用抗酒石酸酸性磷酸酶(TRAP)染色法和骨小坑染色法检测TRAP阳性的破骨细胞量及破骨细胞功能。 结果 在一定范围内,M.CSF在RANKL存在的条件下诱导TRAP阳性破骨细胞成熟,并随M.CSF剂量的增加TRAP阳性细胞数明显增加(r=0.7504,P<0.01);且骨小坑数量也与M.CSF剂量呈明显正相关(r=0.9365,P≤0.0001)。缺乏M.CSF或RANKL均不能诱导TRAP阳性的破骨细胞产生(P=0.0191)。 结论 破骨细胞的增殖、分化及成熟是由M.CSF和RANKL共同调节的。M.CSF是破骨细胞代谢过程中的关键因子,具有促进破骨细胞对骨吸收的作用。M.CSF对骨代谢的主要作用之一是上调破骨系统。
