     Expression of Survivin in two human brain glioma cell lines

    【Abstract】 AIM: To study the expression levels of mRNA and protein of inhibitor of apoptosis protein, survivin, in human brain glioma cell lines BT325 and SHG44 and its significance. METHODS: The expression levels of mRNA and protein of survivin were investigated respectively by reverse transcription polymerase chain reaction (RTPCR) and immunohistochemistry methods in cell lines BT325 and SHG44. RESULTS: The high expression of mRNA and protein of survivin, as well as low expression of splice variant of survivin, survivin2B, was observed in BT325 cells. Protein of survivin was located in cytoplasm of BT325 cells, but mRNA and protein of survivin were not expressed in SHG44 cells. CONCLUSION: Survivin plays an important role in maintaining the biological features of BT325 cells. BT325 and SHG44 cells can be used as ideal cell models respectively for research of gene inhibition and gene transfer of survivin. ......
