【Abstract】 AIM: To develop a HPLC method for the determination of 5hydroxymethylfurfural (5HMF) in prepared rehmannia root. METHODS: The RPHPLC method was used. The analytical column was a ZODBAXC18(4.6 mm×150 mm, 5 μm) and the mobile phase consisted of methanol 0.01 mol/L phosphoric acid aqueous solution (70∶30). The wavelength was set at 284 nm with the Detector and the flowrate 0.8 mL/min, the column temperature 25℃ and the injection volume 20 μL. RESULTS: Over the range of 0.44-2.18 μg, a good linear relation existed between 5HMF and the peak area (r=0.999 9, n=5). The average recovery of the added sample was 99.0% and RSD 0.8% (n=6). CONCLUSION: The proposed method can be used for quantitatively analyzing 5HMF in a convenient and economic way, with high accuracy and good reproducibility. This method for the determination of 5HMF in prepared rehmannia root can be popularized. ......
