视黄酸对人乳腺癌细胞系MCF-7细胞增殖及细胞周期的影响

     【摘要】 目的 观察不同浓度的视黄酸(retinoid acid,RA)对人乳腺癌细胞系MCF-7细胞增殖及细胞周期的影响。方法 以不同浓度RA(10-10~10-6mol/L)处理细胞,采用活细胞计数法观察细胞增殖情况,用氨基安替比林法测定碱性磷酸酶(AKP)活性;通过流式细胞仪技术,测定RA作用后其细胞周期分布。结果 RA对MCF-7细胞的增殖有明显抑制作用,10-7mol/L RA作用5天后活细胞计数为对照组的60%,细胞在G0/G1期停滞;RA还可使MCF-7细胞AKP活性增高,这种作用具有时间和剂量依赖性。结论 RA对MCF-7细胞有明显的增殖抑制作用,细胞在G0/G1期停滞。

    Effects of RA on proliferation and cell cycle of MCF-7 in vitro

    【Abstract】 Objective To study the effects of RA(Retinoid Acid),a synthetic RA,on the cell proliferation and cell cycle of a human breast carcinoma cells.Methods After treated with different concentration of RA(10-10~10-6mol/L),alkaline phosphatase(AKP) activity was determined.The cell cycle progression was examined with flow cytometry.Results RA inhibited the proliferation of MCF-7 cells,the cell number was 60% compared with the control after the treatment with RA(10-7mol/L) for 5 days.AKP activity craised significantly after treatment with RA(10-6mol/L)for 1~5days.Dose-response studies revealed that the AKP activity of MCF-7 cells increased progressively as a function of RA concentrations.Conclusion Glucocortocoids play an important role in the regulation of MCF-7 cell proliferation and differentiation,inducing G0/G1 arrest. ......
