     Response of nigrostriatal neurons to 20 Gy ionizing irradiation and intervening effect of nimodipine

    【Abstract】 AIM: To examine the nigrostriatal neurons in response to ionizing irradiation and the intervening effect of nimodipine so as to provide experimental evidence for the treatment and prevention of ionizing radiation injury or disorders of brain. METHODS: Male BALB/c mice (n=36) were randomly divided into 6 groups: ① blank control, ② singleradiation d 2 group, ③ singleradiation d 7 group, ④ nimodipinetreatment group, ⑤ radiation plus nimodipine d 2 group and ⑥ radiation plus nimodipine d 7 group. Mice were treated with wholebrain irradiation. Immunohistochemistry and semiquantification analysis were utilized to study the distribution and dynamic changes of FOS and nitric oxides synthase (NOS)positive neurons (positive cells/40× field, x±s) in the nigral and striatal regions. RESULTS: FOSimmunostaining showed that compared with that in blank control (16.7±3.7), the expression of FOS in nigral neurons increased markedly in singleradiation d 2 and d 7 groups (28.0±3.6 and 42.0±6.7 respectively, P<0.05-0.01), and more evidently in radiation plus nimodipine groups (36.0±3.6 and 64.0±3.6 in d 2 and d 7 respectively, P<0.05). NOShistochemistry showed that compared with that in blank control (27.4±6.2), NOSpositive neostriatal neurons also went up in singleradiation d 2 and d 7 groups (40.1±8.8 and 39.9±6.8 respectively, P<0.05), whereas they went down in radiation plus nimodipine groups (36.4±6.1 and 31.1±5.2 in d 2 and d 7 respectively, P<0.05). CONCLUSION: The present results suggest that obvious functional changes or injury response occur in the nigrostriatal neurons of animals receiving ionizing radiation, and early administration of nimodipine exerts some intervening effect on the neuronal response to the ionizing irradiation. ......
