【Abstract】 AIM: To determine the effect of the induction of dexamethasone on the proliferation and differentiation phenotype of BMP2 modified rabbit mesenchymal stem cells (rMSCs).METHODS: The rMSCs,treated and untreated with dexamethasone,were transfected with BMP2 gene.The expression of BMP2 was determined by RTPCR and immunocytochemical staining.The changes of the proliferation and differentiation phenotype in the cells were observed by cell growth state,alkaline phosphatase (ALP) activity and osteocalcin (OCN) content.RESULTS: The expression of hBMP2 was confirmed in levels of mRNA and protein.Cell cultures of rMSCs pretreated with dexamethasone showed a higher ALP activity [(13436±884) nkat/L in transfected group versus (10402±783) nkat/L in control group] and higher osteocalcin content [(1468±073) μg/L in transfected group versus (652±121) μg/L in control group].CONCLUSION: Early osteoblastic differentiation induced by dexamethasone can significantly promote BMP2 modified RMSCs osteoblast conversion. ......
