     Early osteoblastic differentiation induced by dexamethasone promotes BMP2 transfected rMSCs osteoblast conversion

    【Abstract】 AIM: To determine the effect of the induction of dexamethasone on the proliferation and differentiation phenotype of BMP2 modified rabbit mesenchymal stem cells (rMSCs).METHODS: The rMSCs,treated and untreated with dexamethasone,were transfected with BMP2 gene.The expression of BMP2 was determined by RTPCR and immunocytochemical staining.The changes of the proliferation and differentiation phenotype in the cells were observed by cell growth state,alkaline phosphatase (ALP) activity and osteocalcin (OCN) content.RESULTS: The expression of hBMP2 was confirmed in levels of mRNA and protein.Cell cultures of rMSCs pretreated with dexamethasone showed a higher ALP activity [(13436±884) nkat/L in transfected group versus (10402±783) nkat/L in control group] and higher osteocalcin content [(1468±073) μg/L in transfected group versus (652±121) μg/L in control group].CONCLUSION: Early osteoblastic differentiation induced by dexamethasone can significantly promote BMP2 modified RMSCs osteoblast conversion. ......
