     Change of mtDNA in liver of subacute aging mouse model induced by high dose Dgalactose injection

    【Abstract】 AIM: To investigate the changes of mtDNA missing in the liver of mice model induced by high dose Dgalactose injection to generate a subacute aging animal model at molecular level. METHODS: Subacute aging animal model was established by Dgalactose(0.1 mg/g.bM)hypodermic injection into scruff, once a day for 45 d. The changes of liver mtDNA missing were detected by PCR. βgalactosidase distribution was observed by βgal staining and level of MDA (malondialdehyde) was determined by fluorospectrophotometry. RESULTS: Compared with those of the control group and young mice group, changes in the liver mtDNA missing were observed in the aging mice model, and the level of MDA increased evidently (P<0.05). The positive blue granules showed by SAβGal staining were found in the aging mice models hepatic cells of lobule circumambience and endothelial cells of brain membrane. CONCLUSION: Subacute aging mice model induced by high dose Dgalactose injection not only has changes in morphology and histopathology but also has deletion of liver mtDNA fragment (3.87 Kb) at molecular level. ......
