氧自由基及NO在内毒素介导的豚鼠耳蜗损伤中的作用

【摘要】 目的:探讨氧自由基及一氧化氮在内毒素介导的豚鼠耳蜗损伤中的作用。方法:选用耳廓反射灵敏的豚鼠36只,随机分为I组:正常对照组(12只);Ⅱ组:生活盐水对照组(12只),双侧听泡各灌注50 μl生理盐水;Ⅲ组:内毒素组(12只),双侧听泡各灌注50 μl LPS (1μg/μl) 。所有动物测试听性脑干反应测听(ABR),并测定耳蜗组织中超氧化物歧化酶( SOD)活力,和耳蜗组织中一氧化氮(NO)含量,同时观察耳蜗组织形态学变化。结果:内毒素组ABRⅠ波、Ⅱ波潜伏期及阈值较其余各组有增大趋势。内毒素组耳蜗组织中SOD活力较其余各组降低 (P<0.05),NO含量较其余各组升高(P<0.05)。内毒素组耳蜗底转内、外毛细胞纤毛出现明显的倒伏、排列紊乱,外毛细胞纤毛损伤从内排到外排逐渐加重,并可见内、外毛细胞纤毛缺失。结论:内毒素导致耳蜗组织中超氧阴离子及NO的大量生成可能是其引起耳蜗组织损伤的部分机制。【关键词 】 内毒素;超氧化物歧酶;一氧化氮;中耳炎
Effect of oxygen free radicals and no on the pathological injury of guinea pig cochlea induced by endotoxin.

【Abstract】 Objective:The aims of this experiment are to study the Effect of Oxygen free radicals and NO on the Cochlea of Guinea Pig. Methods:Thirty-six healthy guinea pigs were randomly divided into 3 groups. GroupⅠ(n=12), the normal control group. Group Ⅱ(n=12), the saline control group. 50μl sterile saline was instilled into each tympanic cavity. Group Ⅲ (n=12), the endotoxin group. 50μl of 1μg/μl solution of LPS was instilled into each tympanic cavity. Inner ear function of the all guinea pigs was assessed by auditory brainstem response ardiometry (ABR). Three of the guinea pigs were used to measure the activities of superoxide dismutase (SOD) in cochlear and five of the guinea pigs were used to measure the concentrations of nitric oxide (NO) in cochlear and two guinea pigs were used to observe morphology features by scanning electron microscope.Results: In group Ⅲ ......
