     Effects of autologous satellite cells transmplantation on cardiac function in acute myocardial infraction

    【Abstract】 AIM: To study the effects of autologous skeleton muscle satellite cells (SC) transplantation on cardiac function in acute myocardial infraction (AMI). METHODS: The proximal potion of left anterior descending coronary artery (LAD) was ligated in young pigs to establish the AMI model and the young pigs were as donors and recipients of autologous SC transplantation. In treatment group, SC were isolated from skeleton muscle, cultureexpanded and labeled with DAPI. The SC were implanted into AMI area through intramyocardium. In control group (n=5), the same volume of cell culture medium was implanted into AMI area through intramyocardium. Gate cardiac blood pool was performed at the end of 1 and 4 weeks after AMI, respectively. The cardiac specimens were harvested for pathological examination at the end of 4 weeks. RESULTS: SC were spindleshaped and adhered on the bottom of the flask after 48hour culture. If calf bovine serum concentration decreased to 20 mL·L-1, SC commenced the fusion process, which resulted in the formation of multinucleated myotubes. New islands and striated muscles were found in treatment group at the site of implantation. The new muscles cell didn't show histological evidence of intercalated discs and centrally located nuclei. Left ventricular ejection fraction (LVEF) increased both in treatment group and control group, but LVEF was higher in treatment group than that in control group [(40±2)% vs (33±2)%, P<0.01], and 1/3 filling fraction were greater in treatment group than that in control group (0.60±0.07 vs 0.54±0.29, P<0.05). CONCLUSION: SC can differentiate into striated muscles under cardiac microenvironment and SC transplantation can improve both systolic and diastolic function of infracted heart. ......
