     HCCX gene expression in hepatocarcinoma and NMR1H spectrum characteristics of correlative marker proteins

    【Abstract】 AIM: To obtain the information expressed by HCCX mRNA in normal liver, parahepatic tissue and hepatocarcinoma with an information probe, catch the correlative protein spectrum characteristics of noncarcinomatous occupancy by the analytical technique of NMR1H spectra, and to discuss their rule and clinical value. METHODS: The obtained gene was labeled as a probe with u-12PDATP.The MTN blot diaphragm was hybridized with 32PHCCX and the function of genetic expression was detected with western blot. Parameter of a chemical shift of 19 kinds of monomeric amino acid (aa)was analyzed by the NMR1H spectrum apparatus. The Hand Book of ProtonNMR Spectra and Data was taken as the reference and analytic conditions were set up. RESULTS: The external clone formation experiment of BEL7402 transfected hepatocarcinoma cells showed that HCCX had a distinct inhibitory effect on the growth of them in vivo with low expression or without expression in hepatocarcinoma tissue and a significant difference as compared with normal tissue (P<0.01); and of the 19 kinds of monomer aa analyzed by NMR1H spectra, 14 were totally accorded with the spectra, 1 was similar, and Trp and His were obtained for the first time respectively. CONCLUSION: HCCX may be the pathogenic gene of hepatocarcinoma and highly homologous HFARP has an inhibitory effect on vascular endothelial cell apoptosis; The conditions of analyzing the correlative marker protein with NMR1H spectra can meet the demands of catching the information from the fingerprint region and provide a new method for the correlative study of physiopathologic mechanism. ......
