复方血竭胶囊对局灶性脑缺血再灌注损伤大鼠血液流变学的影响

     摘要:目的 探讨复方血竭胶囊(CXJC)对局灶性脑缺血再灌注损伤大鼠血液流变学的影响及其意义。方法 雄性SD大鼠40只,随机分为假手术组、缺血再灌注对照组、CXJC低剂量组(0.14 g·kg-1)、CXJC中剂量组(0.56 g·kg-1)和CXJC高剂量组(1.12 g·kg-1),每组8只。采用大鼠大脑中动脉栓塞法制备大鼠脑缺血再灌注模型。缺血2 h再灌注24 h后,经颈总动脉取血,测定全血粘度、全血还原粘度、血浆粘度、红细胞压积、红细胞变形性及红细胞聚集性等血液流变学指标。同时,测定大鼠血清一氧化氮(NO)含量和脑组织谷胱甘肽过氧化物酶(GSHPx)活性。结果 与缺血再灌注对照组相比,CXJC 低、中、高剂量组均能明显降低不同切变率下的全血粘度,降低中、低切变率下全血还原粘度和红细胞压积, 降低红细胞聚集性,并能显著提高红细胞的变形性(P<0.05或P<0.01)。CXJC 0.56、1.12 g·kg-1还可降低缺血再灌注对照组大鼠血浆粘度(P<0.05或P<0.01),降低血清NO含量和增加GSHPx活力(P<0.05,P<0.01)。结论 CXJC通过改善血液流变学和降低脂质过氧化作用而达到抗脑缺血再灌注损伤作用。

    Effects of Compound Xuejie Capsule on the Hemorheology in Focal Cerebral IschemiaReperfusion Rats

    ABSTRACT:Objective To study the effects of Compound Xuejie Capsule (CXJC) on the hemorheology in focal cerebral ischemiareperfusion rats. Methods 40 male SpragueDawley rats were randomly divided into five groups: sham group, ischemiareperfusion group and CXJC groups with lower (0.14 g·kg-1), middle (0.56 g·kg-1) and higher (1.12 g·kg-1) doses. There were 8 rats in each group. The animal models of middle cerebral artery occlusion (MCAO) were established in rats. At 24 h after MCAO, blood was obtained from common carotid artery by polyethylene tube and anticoagulated with heparin 20 KU·L-1, the effects of CXJC on hemorheology including whole blood viscosity, whole blood reduction viscosity, plasma viscosity, haematocrit (Hct), erythrocyte deformation and erythrocyte aggregation were observed. Results Compared with the ischemiareperfusion group, CXJC (0.14, 0.56 and 1.12 g·kg-1) could significantly reduce the increase of whole blood viscosity at highmiddlelow shear rates, decrease the whole blood reduction viscosity at middlelow shear rate, attenuate Hct and erythrocyte aggregation, and increase the erythrocyte deformation in rats (P<0.05 ;P<0.01). CXJC 0.56, 1.12 g·kg-1 could also reduce plasma viscosity (P<0.05;P<0.01). Conclusion CXJC has protective effects on cerebral ischemicreperfusion injury by improving hemorheological parameters and decreasing oxidative stress. ......
