【摘要】 目的 应用全细胞膜片钳研究罗库溴铵、维库溴铵或阿曲库铵对成人型和胎儿型肌肉乙酰胆碱受体的作用。方法 通过脂质体转染法在HEK293细胞表达成人型和胎儿型乙酰胆碱受体。分别研究罗库溴铵、维库溴铵和阿曲库铵各药对两种受体亚型的作用。结果 罗库溴铵、维库溴铵和阿曲库铵呈浓度依赖性抑制乙酰胆碱对成人型和胎儿型乙酰胆碱受体的激动作用。对成人型乙酰胆碱受体抑制程度由大到小依次为维库溴铵>阿曲库铵>罗库溴铵,其对胎儿型乙酰胆碱受体抑制程度由大到小依次为罗库溴铵>维库溴铵>阿曲库铵。结论 不同化学结构肌松药对成人型或胎儿型乙酰胆碱受体的抑制程度不同,而成人型或胎儿型乙酰胆碱受体对一种肌松药的亲和力也有差异。【关键词】 乙酰胆碱受体;肌松药;相互作用;等效线图分析;HEK293细胞
Effects of rocuronium combined with vecuronium and atracurium on adult-and embryonic- muscular nicotinic acetylcholine receptors

【Abstract】 Objective This study was designed to examine the interaction between rocuronium,vecuronium and atracurium on adult-and embryonic-muscular nicotinic acetylcholine receptors with whole cell clamp patch.Methods The adult-and embryonic-nicotinic acetylcholine receptors were heterologously expressed in HEK293 cells and activated to maximal currents by the application of acetylcholine (100μM). Peak currents were recorded in HEK293 cells treated by rocuronium, vecuronium, atracurium individually using whole cell patch clamp technique. Results All compounds tested alone and in combination produced rapid and readily reversible, concentration-dependent inhibition. The potency of vecuronium is the greatest to adult-nicotinic acetylcholine receptor and the potency of rocuronium is the greatest to embryonic-nicotinic acetylcholien receptor, as rocuronium, vecuronium and atracurium applied individually. Conclusion The effects of different muscle relaxants with dissimilar structure were various to the adult-and the embryonic-nicotinic acetylcholine receptors. ......
