银杏天宝对大鼠实验性溃疡性结肠炎IFNγ、IL4的影响

摘要:目的 观察银杏天宝对实验性溃疡性结肠炎大鼠血清、大肠粘膜组织中IFNγ、IL4的影响。方法 采用三硝基苯磺酸(TNBS)灌肠制备大鼠溃疡性结肠炎模型。将动物随机分为空白对照组、三硝基苯磺酸组、三硝基苯磺酸+生理盐水组和三硝基苯磺酸+银杏天宝组,观察肠道大体形态和组织学改变。采用ELISA法测定血清、大肠粘膜中的干扰素γ(IFNγ)和白介素4(IL4)含量。结果 与三硝基苯磺酸组比较,三硝基苯磺酸+银杏天宝组损伤指数明显下降(P<0.01),大肠粘膜和血清IFNγ浓度明显下降(P<0.01),IL4浓度明显升高(P<0.01),IFNγ/IL4比值明显下降(P<0.01)。结论 银杏天宝对溃疡性结肠炎有保护作用,抑制IFNγ、提高IL4生成可能是其作用机制之一。关键词:溃疡性结肠炎;银杏天宝;干扰素γ;白介素4
Effects of Ginkgo Biloba Extract to the Level of IFNγ and IL4 in Experimental Ulcerative Colitis in Rats

ABSTRACT:Objective To investigate the effects of Ginkgo Bilobe extract(EGb) on the level of IFNγ and IL4 in serum and colonic mucosa of rats with experimental ulcerative colitis. Methods The rat ulcerative colitis model was established by using 2.4.6trinitrobenzene sulfonic acid(TNBS).All rats were divided into four groups:normal group TNBS group ,TNBS+NS group and TNBS+EGb group.The macroscopical and histological changes of the colon were evaluated.The IFNγ and IL4 produced by serum and colonic mulosal was analyzed with ELISA. Results In TNBS+EGb group,the macroscopical and histological score was significantly lower than that of TNBS group(P<0.01).In the TNBS+EGb group,a lower level of IFNγ production serum and colonic mucosa(P<0.01) was noticed in comparison with TNBS group.Compared with TNBS group,significantly increased IL4 was noticed(P<0.01) in TNBS+EGb group. The ration of IFNγ/IL4 was lower in TNBS+EGb group compared with TNBS group(P<0.01).Conclusion Ginkgo Biloba extract has protective effect on ulcerative colitis of rat and this effect may be caused by lowering the level of IFNγ and raising the level of IL4 in serum and colonic mucosa. ......
