摘要:目的 观察脂质体介导人纤溶酶原k13基因诱导体外培养人肝癌细胞凋亡的效应。方法 构建人纤溶酶原k13基因真核表达载体pcDNAk13并以阳离子脂质体介导pcDNAk13转染体外培养人肝癌细胞,经Heochst 33258染色后,在荧光显微镜下观察细胞核的形态。结果 转染pcDNAk13组可见致密浓染的凋亡细胞核。结论 脂质体介导人纤溶酶原k13基因能诱导体外培养人肝癌细胞凋亡。关键词:人纤溶酶原k13基因;阳离子脂质体;人肝癌细胞;细胞凋亡
Human Plasminogen k13 gene induces apoptosis in human hepatocellular carcinoma cells by polycationic liposomemediated transfection

Abstract: Objective To investigate the effect on apoptosis of cultured human hepatocellular carcinoma (HCC) cells induced by the human plasminogen kringle 13 (k13) gene by polycationic liposomemediated transfection. Methods recombinant eukaryotic expression vectors pcDNAk13 encoding human plasminogen k13 gene were constructed and transferred to cultured HCC cells by polycationic liposomemediated gene transfer method. After stained with Hoechst 33258 staining solution, and the stained nuclei were observed under a fluorescence microscope. Results condensed bright apoptotic nuclei were readily observed among cells with transfected pcDNAk13. Conclusion human plasminogen k13 gene induces apoptosis in cultured HCC cells by polycationic liposomemediated transfection. ......
