枳椇子对实验性肝纤维化Ⅰ、Ⅲ型胶原的影响

摘 要 目的 研究枳椇子提取物对CCl4诱导的实验性肝纤维化的保护作用。方法 用CCl4制备大鼠肝纤维化模型,免疫组化观察Ⅰ、Ⅲ型胶原蛋白的表达。结果 模型组肝细胞大量坏死,汇管区炎性细胞浸润,假小叶形成。预防组能减轻纤维化时肝细胞的变性坏死及炎性细胞浸润,纤维组织增生较模型组明显减少,呈局限性,假小叶部分消失。预防组Ⅰ、Ⅲ型胶原免疫组化着色减少且浅,呈丝状,与模型组相比程度明显减少。结论 枳椇子能促进肝纤维化时Ⅰ、Ⅲ型胶原的降解。其机理可能与抑制肝星状细胞(hepatic stellate cell,HSC)活化有关。关键词 肝纤维化;枳椇子;胶原
Influnces of Hovenia dulcis Thunb extract on collagen Ⅰ and Ⅲ in hepatic fibrosis

Abstract Objective To investigate the protective effect of Hovenia dulcis Thunb. extract on carbon tetrachloride (CCl4) induced experimental hepatic fibrosis in rats. Methods CCl4 was used to induce liver fibrosis in Wistar Rats. Immunohistochemistry was used to detect the expressions of collagen Ⅰ and Ⅲ. Results In model group, hepatic cells degenerated and necrosed, inflammatory cell infiltrated, and pseudolobuli were formed. In the group treated with Hovenia dulcis Thunb. extract, the degeneration and necrosis of hepatic cells were slight in extent, fibroplasia was much less than that one in the model group, and pseudolobuli partly disappeared. Compared with those in model group, the expressions of collagen I and III in the group treated with Hovenia dulcis Thunb. extract were markedly reduced. Conclusion Hovenia dulcis Thunb. can reduce the expressions of collagen I and III in liver fibrosis. The possible mechanism may be related to the inhibition of the activation of HSC. ......
