摘 要 目的 探讨Aβ25-35对原代培养的海马神经元和隔区神经元毒性作用。方法 采用体外原代培养,用Aβ25-35诱导大鼠海马和隔区神经元损伤,MTT法比较两种神经元的存活率,电镜下的形态学变化、并测定SOD活性的改变。结果 Aβ25-35可使海马和隔区神经元的存活率下降,发生凋亡的形态学改变,对两种神经元的MnSOD的活性无明显影响,但可使海马神经元CuZnSOD活性明显下降。结论 Aβ25-35 具有神经毒性,它对两种神经元的损伤程度相似,无细胞差异性,但其毒性损伤作用可能不同。关键词 淀粉样肽;海马;隔区;神经毒
Comparison of the effect of βamyloid peptide 25-35 on hippocampal and septal neurons in vitroi

Abstract Ojbective To study the difference of Aβ25-35 on neurons between hippocampus and septum. Mdthods employing the method of primary cell culture, neurons survival and SOD were examined ,and amalysed the expression of apoptosisrelated gene bclxl by Western blot. Result Aβ25-35 might reduce the survival of hippocampal and septal neurons, increased both activity of CuZnSOD and expression of Bclxl, but decreased the activity of MnSOD of hippocampal neurons, and had on obvious effect on septal neurons. Conclusion Aβ25-35 had the same effect on survival and CuZnSOD but MnSOD between the hippocampus and septum. The cytoxic mechanisms of Aβ25-35 on hippocampus and septus might be different. ......
