A β25-35对海马和隔区胆碱能神经元毒性的比较

     摘 要 目的 探讨Aβ25-35对原代培养的海马神经元和隔区神经元毒性作用。方法 采用体外原代培养,用Aβ25-35诱导大鼠海马和隔区神经元损伤,MTT法比较两种神经元的存活率,电镜下的形态学变化、并测定SOD活性的改变。结果 Aβ25-35可使海马和隔区神经元的存活率下降,发生凋亡的形态学改变,对两种神经元的MnSOD的活性无明显影响,但可使海马神经元CuZnSOD活性明显下降。结论 Aβ25-35 具有神经毒性,它对两种神经元的损伤程度相似,无细胞差异性,但其毒性损伤作用可能不同。

    Comparison of the effect of βamyloid peptide 25-35 on hippocampal and septal neurons in vitroi

    Abstract Ojbective To study the difference of Aβ25-35 on neurons between hippocampus and septum. Mdthods employing the method of primary cell culture, neurons survival and SOD were examined ,and amalysed the expression of apoptosisrelated gene bclxl by Western blot. Result Aβ25-35 might reduce the survival of hippocampal and septal neurons, increased both activity of CuZnSOD and expression of Bclxl, but decreased the activity of MnSOD of hippocampal neurons, and had on obvious effect on septal neurons. Conclusion Aβ25-35 had the same effect on survival and CuZnSOD but MnSOD between the hippocampus and septum. The cytoxic mechanisms of Aβ25-35 on hippocampus and septus might be different. ......
