海藻糖对液态低温保存血小板保护作用的实验研究

     [摘要]

    目的:研究海藻糖对液态低温(4℃)保存血小板的保护作用。方法:采集兔心脏血,按常规方法制备浓缩血小板(CP),在CP中加入不同浓度的海藻糖,4℃储存,用51Cr铬酸钠进行标记后自体回输入兔体内,于回输后1、24、48、72h抽取兔耳动脉血,测定放射性计数率,计算24、48、72h血小板存活率,同时设常温对照组及单纯4℃储存的低温对照组进行比较。结果:加入不同浓度海藻糖各储存组的血小板寿命明显长于低温对照组,并且随着海藻糖浓度的增加而延长。结论:海藻糖可延长4℃液态保存血小板的寿命,对血小板具有保护作用。

    Experimental research on the protective effect of trehalose on liquid platelets stored at low temperature

    Abstract:Objective To study the protection of trehalose for liquid rabbit platelets stored at 4℃ traced by 51Cr. Methods Cardiac blood was taken and made into concentrated platelet(CP).CP were stored at different conditions,such as low temperature and low temperature+different dose of trehalose for 24h.Platelets were transfused into rabbits’ bodies after being labelled with 51Cr and platelet lifespan of 24,48,72h was calculated. Results After trehalose was added into the platelets stored at 4℃,the lifespan was obviously longer than that of platelets stored at low temperature. And with the dose increasing,the lifespan of platelets wes prolonged obviously till the dose was more than 50mg·ml-1. Conclusion Trehalose ......
