八角莲不同部位鬼臼毒素分布的毛细管区带电泳研究

     摘 要

    建立了一个简单灵敏的分离测定鬼臼毒素的高效毛细管区带电泳方法,并用该方法研究了八角莲的不同部位中鬼臼毒素的分布情况。结果表明,在未涂层石英毛细管(57 cm×75 μm i.d.,有效长度50 cm)中,以25 mmol/L的硼砂(pH 10.8)缓冲溶液为电解质,鬼臼毒素可与八角莲不同部位中的其它组分得到有效分离。方法对鬼臼毒素的测定范围为1.5 ~ 171.0 mg/L,相对迁移时间和峰面积的RSD值分别为1.50%和0.82%(n=8)。对八角莲的根、茎、叶、须乙醇提取物进行单独测定,发现鬼臼毒素在八角莲不同部位中的分布是不均匀的,其中根部最多,而叶中最少。

    The Distribution of Podophyllotoxin in the Different Parts of Dysosma Versipellis (Hance) M. Cheng by Capillary Zone Electrophoresis

    Abstract A simple and sensitive capillary zone electrophoresis (CZE) method for the determination of podophyllotoxin (PPT) has been developed, and the distribution of podophyllotoxin in the different part of Dysosma versipellis (Hance) M. Cheng was studied by the proposed method. The results indicated that podophyllotoxin could be separated availably from the other constituents in the different part of Dysosma versipellis (Hance) M. Cheng in an uncoated fusedsilica capillary (57 cm×75 μm i.d. with the effective length of 50 cm) using 25 mmol/L borate with pH 10.8 as running buffer. The linear range for the determination of podophyllotoxin was 1.5-171.0 mg/L with the relative standard deviations (n=8) for the relative migration time and peak area of 1.5% and 0.8%, respectively. The results obtained from the determination of podophyllotoxin in the ethanolic extracts of the roots, stems, leaves and fibrils of Dysosma versipellis (Hance) M. Cheng pointed out that the distribution of podophyllotoxin in the different part of Dysosma versipellis (Hance) M. Cheng was not equal.
