摘 要:[目的]观察神经干细胞缺氧后的内源性促红细胞生成素(erythropoietin,EPO)表达规律,初步探讨缺氧预处理诱导神经保护作用的机制。[方法]测定大鼠神经干细胞缺氧培养4 h后不同时间的内源性EPO表达情况,以及预处理后不同时间,再次遭受致死剂量缺氧伤害后的EPO表达情况。[结果]缺氧时间为4 h,EPO蛋白在处理后即刻出现表达,4 h达高峰,8 h仍有部分表达。再次遭受致死剂量缺氧伤害后,与预处理4 h组相比,预处理24 h组细胞的EPO表达量明显增多。[结论]缺氧预处理能够提高神经干细胞应对伤害刺激时内源性EPO的表达,并降低再次缺氧时诱导EPO表达的阈值。关键词:缺氧; 预处理; 干细胞; 促红细胞生成素

Abstract:[Objective]To investigate the endogenous erythropoietin expression of neural stem cells(NSCs) of rats after hypoxia.[Method]We used a rat NSCs culture model. EPO expression after 4 hours of hypoxia was studied. It was examined again following 12 hours of hypoxic lethal insult which was applied various times after hypoxic preconditioning. EPO expression was studied by western blotting.[Result]Following the preconditioning, expression of EPO increased immediately and reached a peak 4 hours later.It lasted till 8 hours later.While applied the lethal hypoxia,EPO expression of cells in preconditioning 24 hours group were was significantly increased than that in preconditioning 4 hours group.[Conclusion]The hypoxic preconditioning can lead an increased endogenous EPO expression and decrease the threshold of EPO expression against subsequent lethal stimulation during a certain period. ......
