一氧化氮与哮喘气道炎症实验研究

     [摘要]目的评价一氧化氮(Nitric Oxide,NO)在哮喘气道炎症防治中的作用。方法用两种免疫学方法(酶联免疫吸附试验和免疫组化ABC方法)和生理学方法测定了哮喘组和正常对照组豚鼠血浆和肺泡灌洗液(BALF)中NO水平和肺组织上皮细胞中NOS(一氧化氮合成酶)、ICAM1(细胞间粘附分子1)表达及肺功能改变。结果与对照组比,①哮喘豚鼠血浆可溶性ICAM1水平上升,BALF中NO水平上升(P<005);②肺组织NOS、ICAM1表达水平上调(P<005);③肺潮气量和动态肺顺应性下降,气道阻力上升(P<005)。结论NO介导了哮喘气道炎症。

    Effects of NO on the airway inflammation of bronchial asthma

    [Abstract]ObjectiveTo investigate the effects of NO on the airway inflammation of bronchial asthma.MethodsThe guinea pigs were divided into 2 groups: asthma group and control group. The following tests were performed on 2 groups. The sICAM1 value in plasma and NO value both in plasma and BALF were measured by ELISA. The expressions of ICAM1 and NOS on the bronchial epithelial cells were measured by immunohistochemistry staining. The lung fuctions (VT, Cdyn, RL) were tested. ResultsCompared to the control group, ①Apart from the value of NO in plasma level of asthmatic animals, the values of sICAM1 in plasma and NO value in the BALF were elevated; ②The expressions of ICAM1 and NOS on the bronchial epithelial cells were upregulated; ③The lung compared funtions (VT, Cdyn, RL) were impaired. ConclusionsAll the results above suggests NO is involved in the inflammation of bronchial asthma. ......
