血管内皮细胞生长因子和血管发生素1及其受体在斑马鱼早期胚胎发育过程中的表达

摘要:目的探讨血管内皮细胞生长因子和血管发生素1及其受体在斑马鱼早期胚胎发育过程中的表达,为构建药物筛选的在体模型提供依据。方法采用多聚酶链式反应,扩增了长约280bp的斑马鱼血管发生素1 cDNA部分序列,作为探针,以原位杂交法测定了血管内皮细胞生长因子和血管发生素1及其相应受体flk1和Tie2的表达。结果从受精卵胚胎发育第12小时可见血管内皮细胞生长因子和血管发生素1及其相应受体flk1和Tie2的表达;在受精卵胚胎发育的第16小时,血管内皮细胞生长因子和血管发生素1主要在头部和尾部胚芽表达;第24小时在眼球、视窝、中脑、后脑的腹前侧和肌节中表达,Tie2和Flk1主要在肌间背部动脉血管、躯干新生血管芽和轴向静脉中表达。结论血管内皮细胞生长因子和血管发生素-1及其受体在斑马鱼胚胎发育早期的血管发生过程中可能相互作用,并协调血管发生过程。斑马鱼是研究胚胎发育早期微血管发生和血管发生的极佳动物模型,可用于促血管发生或抑制血管发生药物筛选。关键词:斑马鱼; 血管发生素1; 血管内皮生长因子; 受体
Expressions of Angiopoietin1 and Its Receptors During Early Development in Zebrafish

Abstract:ObjectiveTo investigate the expression of vascular endothelial growth factor (VEGF) and angiopoietin1 (Ang1), and their receptors Flk1 and Tie2 during angiogenesis and vasculogenesis in early embryo development of zebrafish. MethodsUsing homology-based polymerasechain reaction, a 280 bp of partial cDNA of zebrafish (zfAng1) was isolated. The expressions of Ang1, Tie2, VEGF and Flk1 in zebrafish were examined during up to 24 h embryonic development by whole mount in situ hybridization. ResultsThere were no detectable expressions of these genes up to 10 h postfertilization. However, there were detectable expressions of these genes at 12 h. At 16 h, Ang1 and Tie2 were expressed mainly in head and tail bud. At 24 h, Ang1 was expressed mainly in the lens, optic cup, mesencephalon, ventral part of hindbrain, and myotomes in the somites, while zfTie2 was mainly expressed in intersomitic vessels including dorsal aorta and axial veins, and sprouting vessels from the dorsal aorta in the trunk. VEGF was expressed mainly in the lens, optic cup, mesencephalon and myotomes in the somites, while zfFlk1 was mainly expressed in intersomitic vessels including dorsal aorta and axial veins, and sprouting vessels from the dorsal aorta in the trunk. ConclusionAng1, VEGF, Tie2 and Flk1, may act cooperatively and interactively for vascular development in zebrafish during early development. Present results also suggested that zebrafish as a quality model for study on vasculogenesis and angiogenesis during embryonic development and screening for angiogenic enhancer or inhibitors. ......
