HL60细胞冻存复苏生物学特性对比分析

     摘 要:目的 探讨不同冻存复苏条件对白血病细胞株HL60生物学特性的影响,并优化培养方法与条件。方法 对比不同浓度DMSO冻存条件、不同复苏方法对复苏后细胞生物学特性的影响,并确定培养液血清最佳浓度。结果 采用5%DMSO冻存防护效果好于其他组,改良后细胞的复苏存活率明显高于传统复苏方法。结论 以5%DMSO作为HL60细胞冻存保护剂并采用改良复苏方法,可使细胞保持最佳生物学特性,12%血清为HL60细胞常规培养的最适浓度。

    Comparison of biologicphenotypes of cultured HL60 after cryopreservation

    Abstract:Objective To probe the influence of different frozen and recovery conditions on the biology characteristic of HL60, and optimized the culture technique. Methods Compare resuscitation effect of different freezing solutions with DMSO at different concentrations and of different resuscitation methods by the cell recovery rates which were respectively examined by trypan blue exclusion staining after thawingConfirm the best serum content by observing the grow circus of HL60 in different serum content culturemedium. Results 5% DMSO was the favorable concentration for cryopreservation of HL60; The livability of dissolve in 40℃advance plus 37℃ resuscitation method is evidently higher than the tradition 37℃ resuscitation method.Cells grow well in RPMI1640 that contain12% new cattle serum. Conclusion We can get the better biologicphenotypes of HL60 after cryopreservation by using 5%DMSO as cryoprotectant,taking the improved resuscitation method and growing in RPMI1640 that contain 12% newl cattle serums. ......
