MMP9和PTEN在乳腺癌中的表达及其临床意义

     摘要:目的: 探讨基质金属蛋白酶9(MMP9)和PTEN在乳腺癌中的表达及其临床意义。方法:运用免疫组化Envision方法检测56例乳腺癌和22例乳腺良性病变中(乳腺腺病和乳腺腺瘤各11例)MMP9和PTEN的表达情况, 并分析其与临床病理参数的关系。结果:(1)MMP9在乳腺癌中的阳性率为69.64%(39/56),在乳腺良性病变中的阳性率为45.45%(10/22);PTEN在乳腺癌中高表达率为46.43%(26/56),在乳腺良性病变中为86.36%(19/22),两组差异有统计学意义(P<0.05)。(2)MMP9的表达与组织学分级和腋窝淋巴结转移呈正相关。PTEN蛋白的表达与腋窝淋巴结转移呈负相关,而与组织学分级无关(P>0.05)。结论:(1)MMP9参与乳腺癌的发生发展过程,可作为评估乳腺癌浸润和转移的重要生物学指标。(2)乳腺癌中存在PTEN蛋白表达异常,可能与乳腺癌的发生、发展和预后有关。

    Expression and clinical significance of MMP9 and PTEN in the tissues of breast cancer

    Abstract: Objective: To study expression of matrix metalloproteinase9 (MMP9), and phosphatase and tensin homolog deleted from chromosome 10 (PTEN) in breast cancer and evaluate its significance to clinicopathology of this tumor invasion and metastasis. Methods: The immunohistochemical staining method of Envision was used to evaluate the expression of MMP9 and PTEN in 56 cases of breast cancer and 22 cases of benign breast diseases and their correlations to clinicopathologic features were analyzed. Results: (1)The positive incidence of MMP9 expression in 56 cases of breast cancer were 69.64%(39/56), which was improved compared with the expression in benign breast diseases were 45.45%(10/22) (P<0.05). The higher level expression of PTEN in 56 cases of breast cancer with the rate 46.43%(26/56), which was reduced compared with the expression in benign breast diseases were 86.36% (P<0.05). (2) The expression of MMP9 was positively correlated to axillary lymph node metastasis and pathological stage. The expression of PTEN was negatively correlated to axillary lymph node metastasis. On the contrary, there was no significant correlation between PTEN expression and pathological stage (P>0.05). Conclusions: (1) MMP9 was associated in breast cancer and could serve as an important index in the aspect of invasiveness and metastasis in breast cancer. (2) It showed that tumor suppressor PTEN gene was abnormally expression in breast cancer.And PTEN is probably related to carcinogenesis and development of breast cancer. ......
