肥大细胞膜稳定剂对大鼠重症急性胰腺炎肠组织保护作用的实验研究

     【摘要】目的:探讨肥大细胞(MC)稳定剂酮替芬对重症急性胰腺炎(SAP)大鼠肠道组织的保护作用。方法:SAP大鼠模型前或模型后30min给予酮替芬,模型后3h及6h取大鼠末端回肠组织行HE染色观察病理变化。甲苯胺蓝及免疫组化染色计算MC数目,测定髓过氧化物酶(MPO)活性了解中性粒细胞(PMN)聚集程度,Western Blot法检测类胰蛋白酶表达。结果:酮替芬应用组肠组织病理损害较SAP组明显减轻;酮替芬预处理组3h及6h甲苯胺蓝染色每高倍视野MC计数分别为1.21及1.08,相应时段SAP组MC计数为3.52及1.38,两组3h MC计数差异有统计学意义,6h MC计数差异无统计学意义;酮替芬早期治疗组3h及6h MC计数为1.19及1.03,与酮替芬预处理组同时段比较差异无统计学意义;各组免疫组化染色MC计数亦呈上述趋势;酮替芬处理组MPO活性较SAP组明显降低;酮替芬处理组类胰蛋白酶表达较SAP组明显下降。结论:预防性或SAP早期应用酮替芬,可减轻SAP肠组织的损害。

    Experimental study of protective effects of mast cell stabilizer on ileum of rat with severe acute pancreatitis

    【ABSTRACT】Objective:To investigate the protective role of mast cells stabilizer on ileum of rats with severe acute pancreatitis(SAP).Methods:Ketotifen was intraperitoneal injected 30 minutes before or after SAP model,after 3 hours and 6 hours,terminal ileum was removed to make HE staining,to count the number of MC by the method of toluidine blue staining and immunohistochemistry staining,to measure the level of MPO and activation level of tryptase by western blot,and compare these parameters with those of SAP rats without ketotifen treatment.Results:The pathologic damage of ketotifen treatment group was lighter than SAP groups.The MC numbers per high visual field of 3h and 6h of ketotifen pretreatment group were 1.21 and 1.08,and the MC numbers of SAP group 3.52 and 1.38,there was significant difference between 3h groups,but no significant difference between 6h groups.There was no significant difference between the ketotifen pretreatment group and the early treatment group;the data of MC number by immunohistochemistry staining has the similar trend,and the activation level of MPO of ketitofen group was lower than SAP group,And the expression of tryptase in ketotifen group decreased compared with that of SAP group.Conclusion:MCs are activated in the early period of SAP,and induce damage of ileum.Pretreatment or early using of ketotifen can decrease the severity of ileum in SAP. ......
