HPLC-UV法测定人血浆中头孢羟氨苄含量

     【摘要】 目的 建立用内标法测定人血浆中头孢羟氨苄浓度的HPLC-UV方法。方法 用10%高氯酸直接沉淀血浆蛋白,以对氨基苯甲酸为内标,测定血浆中头孢羟氨苄的浓度。结果 血浆中杂质不干扰样品峰和内标峰,20.0、2.0、0.2μg/ml的回收率分别为98.1%,98.7%, 91.1%;批内精密度均小于8.0%(高、中、低浓度分别为3.8%,5.6%,7.3%),批间精密度均小于10%(高、中、低浓度分别为 4.1%,6.5%,9.7%),最低检测限为0.1μg/ml,线性范围为0.2~30.00μg/ml;血浆中头孢羟氨苄冻融是稳定的;精密度也符合要求。结论 该方法稳定且精密度好,准确可靠,适用于头孢羟氨苄血浆浓度测定。

    【Abstract】 Objective To establish the determination method of cefadroxil in healthy volunteers plasma. Methods Plasma protein was precipitated by perchloric acid (10%), and paraaminobenzoic acid was used as internal standard. Plasma concentrations of cefadroxil and internal standard were determined by HPLC-UV.Results The chromatographic peak of cefadroxil and internal standard was not interfered by foreign matter. Recovery rate of 20.0、2.0、0.2μg/ml was 98.1%, 98.7%, 91.1%, respectively. Precision in or between batch was less than 8.0% and 10%, respectively. Lowest detectable limit was 0.1μg/ml and linear range was 0.2~30.00μg/ml. The freeze thawing of plasma was stable.Conclusion The method was reliable and stable, it was suitable for determination the concentration of cefadroxil in plasma. ......
