乳腺癌患者RASSF1A基因启动子区甲基化及临床意义

【摘要】 目的 探讨乳腺癌组织中RASSF1A(RAS association domain family 1A)基因启动子区CPG岛甲基化状态及临床意义。方法 用甲基化特异性PCR(MSP)检测40份乳腺癌组织和24份正常乳腺组织中RASSF1A基因启动子区甲基化状态。结果 乳腺癌组织中RASSF1A基因启动子区CPG岛甲基化率为45.0%(18/40),与正常乳腺组织比较差异有显著性(2/24)(P<0.05),但与肿瘤病理类型、患者年龄、有无淋巴结转移的差异无显著性(P>0.05)。结论 RASSF1A基因启动子区在乳腺癌中发生异常甲基化,为乳腺癌的候选基因。【关键词】 乳腺癌; RASSF1A基因;基因甲基化;甲基化特异性PCR
Promoter methylation of RASSF1A gene in breast cancer patients and its clinical significance

【Abstract】 Objective To investigate the methylation status of the CPG islands in the promoter regions of RASSF1A gene and to analyze its clinical significance in breast cancer patients.Methods Methylation status of RASSF1A promoter was detected by MSP in 40 cases of breast cancer tissues and 24 cases of matched benign tissues. Results The frequency of promoter methylation of RASSF1A gene in tumor tissues was 45.0%(18/40),which was significantly higher than in benign tissues(8.3%,2/24)(P<0.05).But no significant association of abnormal methylation was identified with histological type ,the patient’s age ,clinical features such as lymph node metastasis. Conclusion Our findings showed that there was hypermethylation of CPG islands in the promoter area of RASSF1A in breast cancer .So RASSF1A gene may be one of the new candidates of tumour suppressor genes. ......
