【Abstract】 AIM: To explore the effect of 17β estradiol (E2)on apoptosis and membranetype matrix metalloproteinase 1(MT1MMP) mRNA expression in endothelial cell line EVC304 under the high glucose condition 30 mmol/L. METHODS: The proliferation of EVC304, which is treated by high glucose(30 mmol/L) and/or E2 of different concentrations respectively, was measured by MTT assays. Then the apoptosis of ECV304 was detected by flow cytometry(FCM) and DNA ladder assays. Finally, MT1MMP mRNA expression was measured by semiquantitative RTPCR. RESULTS: After ECV304 had been treated by high glucose(30 mmol/L) ,its proliferation and mRNA expression of MT1MMP was inhibited significantly(P<0.01). However, the tendency fall back after E2 had been administrated(P<0.01). Furthermore, this effect of E2 reached a peak at its concentration from 0.01 to 0.10 mmol/L. The apoptosis was only detected in cells which had been administrated by high glucose and E2 at low (0.001 nmol/L) and high (1.0 nmol/L) concentration by DNA Ladder assays. CONCLUSION: High glucose has a strong toxicity on vascular endothelial cells. Moderate level of E2 has a protective effect on vascular endothelial cells. E2 can enhance MT1MMP expression of endothelial cells under high glucose condition. ......
