     Change of Na+, K+ATPase α isoforms in rat neurons damaged by global cerebral ischemia/reperfusion

    【Abstract】 AIM: To investigate the change of Na+, K+ATPase α isoforms in rat neurons damaged by global cerebral ischemia/reperfusion. METHODS: The globral cerebral ischemia (15 min) in rats were induced by Pulsinelli 4VO method and the rats were reperfused for 30 min. With RTPCR and immunohistochemical assay, the expression of the Na+, K+ATPase α isoforms was detected in rat hippocampal and cortical neurons following ischemia/reperfusion. RESULTS: The pyramidal cells during ischemia/reperfusion displayed notable edema. The α1, α2 or α3 isoform of Na+, K+ATPase distributed in a tissuespecific fashion in neurons of hippocampus and cortex, and α3 isoform was more than α2 and α1 isoforms in hippocampus, while there was no significant difference in cortex. The expressions of α1 and α3 isoforms in the rats suffering ischemia/reperfusion were significantly decreased in hippocampus (P<0.05 or 0.01). However, the expressions of α1 and α3 in cortex did not change during 15min ischemia and declined in 30min reperfusion (P<0.05). In both ischemia and reperfusion, the mRNA expressions of α1 and α3 isoforms at mRNA level had no significant change as compared with shamoperated group. CONCLUSION: The change of the expressions of α1 and α3 isoforms may involve in global cerebral ischemia/reperfusioninjury. ......
