砷剂联合化疗药物治疗MDR人肝癌细胞初步研究

     [摘要] 目的 通过实验观察砷剂联合化疗药物对人肝癌MDR细胞的治疗和逆转作用,为临床改善肝癌疗效提供理论依据。方法 按砷剂单用或联合化疗药物作用人肝癌细胞QGY-7703/cDDP和HepG2/ADM进行分组,采用光镜和电镜观察细胞形态结构及超微结构的改变,台盼蓝染色活细胞计数测定细胞增殖能力,MTT检测细胞毒作用,FCM进行细胞周期分布分析。结果 肿瘤细胞药物作用后,电镜观察到细胞凋亡和凋亡小体,肿瘤细胞倍增时间延长,生长曲线下移,细胞毒作用明显,G0/G1期细胞比例下降,G2/M期细胞比例增加。结论 砷剂单用和联合化疗对MDR肝癌细胞生长具有抑制作用,同时能够部分逆转人肝癌细胞MDR。

    Study on combination of arsenic trioxide and chemical therapy in treatment of MDR of human hepatocellular carcinoma

    [Abstract] Objective To study combination of arsenic trioxide and chemical therapy in treatment of MDR of human hepatocellular carcinoma.Methods The human hepatocellular carcinoma HepG2/ADM and QGY-7703/cDDP were made into every grades by single and combined arsenic trioxide. HepG2 and QGY-7703 were control grades. Morphologic studies were performed with optical microscopy and transmission electron microscopy after the human hepatocellular carcinoma cells were exposed to various drugs. Cell viability was evaluated by the trypan blue exclusion test. Killing effects and proliferative activities were assessed by MTT assay. Cell cycle analysis was performed by flow cytometry (FCM).Results Apoptotic cell and apoptotic body were observed by transmission electron microscopy in carcinoma cells were exposed to various drugs. At one time, compared to control group, the tumor doubling time were significantly extended, and tumor growth curves were significantly removed down,cell killing effects were significant,cell cycle analysis showed cellular inverse proportions of G0/G1 fall and G2/M elevated.Conclusion The single and combined arsenic trioxide could restrain the growth of the human hepatocellular carcinoma of MDR, furthermore reverse partially the MDR. ......
