活化造血免疫细胞回输对高剂量化疗恶性肿瘤患者的免疫保护作用

     摘要目的：探讨活化的造血免疫细胞回输对高剂量化疗患者的免疫保护作用。方法：经过3～6个疗程的常规化疗或常规放疗未完全缓解或缓解后复发的恶性肿瘤26例，随机分为造血免疫细胞活化组(简称活化组)16例和未活化组10例。活化组患者化疗前用GMCSF动员5 d后，从外周血分离出单个核细胞，用GMCSF、抗肿瘤抗T细胞双功能抗体、IL2在体外活化和扩增粒系祖细胞和免疫细胞，在亚去髓性化疗后回输给患者。结果：(1)与未活化组相比，活化组患者的中性粒细胞绝对数(ANC)从<0.1×109/L升至>0.5×109/L的时间缩短6.1 d；(2)体外培养CFU-GM增加9.8倍，有核细胞数增加3.4倍；(3)T淋巴细胞对抵抗NK活性的肺癌细胞株PC 84045的杀伤率从10%提高至55%，对K 562细胞杀伤率从28%提高至46%。结论：用活化的造血免疫细胞回输能加快高剂量化疗后造血和免疫功能的重建。
