丹参在大鼠恶性梗阻性黄疸时对肿瘤细胞凋亡的作用

    【Abstract】 AIM: To establish malignant biliary obstruction models in SD rats, and to study the effects of Danshen injection on the growth of hepatocellular carcinoma, morphological change and PCNA expression and apoptosis index in carcinoma and pericarcinoma tissues. METHODS: ① Malignant biliary obstruction model in SD rats was made by vaccination of transplanted tumor in liver nearby the porta hepatis with Walker256 hepatocarcinoma cell line, which infiltrated proximal common bile duct to cause its narrowness and obstruction. ② The SD rats were divided into 4 groups that were treated ip by 0.9% normal saline (NS) (n=16), by inosine+vitamin C (n=26), by Danshen (n=34) and by 5FU (n=30) respectively. ③ The diameter of transplanted tumor, morphological change and the expression of PCNA and apoptosis index in carcinoma and pericarcinoma tissues were observed. ④ Statistical analysis was made in the experimental data. RESULTS: In Danshen group as compared with NS group and inosine+vitamin C group, the average size of hepatocarcinoma was reduced significantly, the tumor inhibitory rate and the metastasisinhibitory rate were higher obviously (P<0.01), PCNA expression was lower (P<0.01), apoptosis index was higher (P<0.01); while compared with 5FU group, the average size was significantly larger (P<0.05) but the metastasisinhibitory rate was lower evidently (P<0.05), apoptosis index was higher in carcinoma tissues (P<0.05). CONCLUSION: Danshen shows a protective effect against morphological change in SD rat models of malignant biliary obstruction, and exerts an inhibitory effect on the growth of hepatocarcinoma through suppressing the proliferation, increasing the apoptosis of the hepatocarcinoma cells. ......
