Aβ2535海马注射对大鼠神经细胞中NFκB活性的影响

【摘要】 目的 在体条件下观察β淀粉样蛋白(βamyloid,Aβ)对核因子κB(nuclear factor κB,NFκB)活性的影响,探讨二者之间的关系。 方法 以Aβ2535及生理盐水做海马立体定向注射后,采用水迷宫、HE染色及免疫组织化学方法,观察实验动物学习记忆改变、脑组织病理形态变化及大鼠海马部位NFκB活性的变化。 结果 Aβ组和生理盐水对照组通过水迷宫的潜伏期分别为(5993±2343)s和(856±481)s,差异显著(P<001),而Aβ组海马各区及齿状回可见大量的NFκB阳性细胞,以齿状回和CA1区明显,与对照组相比,差异显著。 结论 在体条件下Aβ能激活NFκB,提示NFκB的活化与Aβ的神经细胞毒性有关。【主题词】 淀粉样β蛋白; NFκB; 阿尔顿海默病
Activation of NFκB after βamyloid injection into hippocampus of rats

【Abstract】 Objective To investigate the effects of βamyloid on NFκB in vivo. Methods One week after sterotaxis injection of Aβ2535 and normal saline into CA1 subregion of hippocampus, the changes of their memory recovery, the hippocampus pathological abnormalities and the activity of NFκB were evaluated with Morris test, hematoxylineosin staining and immunohistochemical assays. Resukts The latency of Aβ group(5993±2343 s) was significantly different from that of saline group(856±481 s), while a large number of NFκB positive cells were observed in hippocampus of Aβgroup, especially in CA1 subregion and dentate gyrus, which were different from those of saline group. Conckusions Aβ could induce the activation of NFκB, which is related to the neurotoxity of Aβ. ......
