FL,TPO对人脐带血CD34+细胞体外增殖的影响

[摘要] [目的] 研究Flt-3配体(FL)及血小板生成素(TPO)在无血清培养体系中对人脐带血CD34+细胞体外增殖的影响. [方法] 采用无血清液体培养法,分别用FL,TPO,FL加TPO作用于体外短期培养脐带血CD34+细胞,利用其计数及集落分析方法评价增殖效果. [结果] 分别用FL,TPO,FL加TPO刺激增殖后,人脐带血细胞总数分别增加2.50,5.00,10.10倍,FL加TPO组细胞总数明显高于FL,TPO组;CD34+细胞分别增殖0.90,1.26,1.61倍;集落总数分别增加2.07,2.27,7.37倍,造血祖细胞CFU-GM产率分别提高3.53,2.40,9.95倍,FL加TPO组产率高于其他组,BFU-E产率分别提高1.45,2.25.6.90倍. [结论] 在短期无血清液体培养体系中,FL,TPO对人脐带血CD34+细胞均具有增殖作用,且两者具有协同作用.[关键词] 配位体;血小板生成素;胎血;细胞学技术
