[摘要] [目的] 研究Flt-3配体(FL)及血小板生成素(TPO)在无血清培养体系中对人脐带血CD34+细胞体外增殖的影响. [方法] 采用无血清液体培养法，分别用FL，TPO，FL加TPO作用于体外短期培养脐带血CD34+细胞，利用其计数及集落分析方法评价增殖效果. [结果] 分别用FL，TPO，FL加TPO刺激增殖后，人脐带血细胞总数分别增加2.50，5.00，10.10倍，FL加TPO组细胞总数明显高于FL，TPO组;CD34+细胞分别增殖0.90，1.26，1.61倍;集落总数分别增加2.07，2.27，7.37倍，造血祖细胞CFU-GM产率分别提高3.53，2.40，9.95倍，FL加TPO组产率高于其他组，BFU-E产率分别提高1.45，2.25.6.90倍. [结论] 在短期无血清液体培养体系中，FL，TPO对人脐带血CD34+细胞均具有增殖作用，且两者具有协同作用.
