【Abstract】 AIM: To study the process and influencing factors of synthesizing the melanocyte stimulating hormone(MSH) peptide by Fmoc solidphase synthesis. METHODS: With Fmoc to protect the αamino acid, TBTU/HOBT/DIEA were used as coupling reagents to synthesize the peptide of MSH. The synthesized peptide was purified by the chromatograph and identified by mass spectrograph. RESULTS: The peptide yield was 63%.The synthesized peptide purity reached to 98.67% after being purified by RPHPLC. The molecular mass of the synthesized peptide was 1663.88 ku by MALDIMS and it was consistent with the theoretic molecular mass of 1664.08 ku. CONCLUTION: This method of synthesis is rapid,simple and effective for the peptide MSH, and its fit for the largescale production.
