褪黑激素对严重烧伤大鼠血管通透性的影响

[摘要]目的 观察褪黑激素对严重烧伤大鼠血管通透性的影响。方法 30只大鼠随机分为假烫对照组、烧伤对照组和褪黑激素组,每组10只。将麻醉动物背部皮肤置于沸水或37 ℃水中15 s,造成30% Ⅲ度烫伤或假烫伤。假烫对照组和烧伤对照组术后立即腹腔注射溶剂(体积分数0.01的乙醇生理盐水),而褪黑激素组腹腔注射10 mg/kg褪黑激素。烧伤后6 h取血和皮肤、肝、肾、肺组织进行测定。结果 严重烧伤可导致血细胞比容(HCT)增加( F=15.19,q=7.79,P <0.01),血浆总蛋白(TP)浓度下降( F=5.95,q=4.50,P <0.05),血浆丙二醛(MDA)水平升高( F=8.06,q=5.57,P <0.01)。以上改变可部分被褪黑激素所拮抗( F=5.95~15.19,q=3.44~3.81, P < 0.05、0.01)。烧伤后肾、肺和皮肤组织MDA水平均明显升高( F=11.18~66.46,q=3.95~16.25,P < 0.01 )。褪黑激素治疗使肾和肺组织MDA降至假烫对照组水平( F=11.18、21.30,q=2.42~7.43,P <0.01),使皮肤MDA降低25.4%( F=66.46,q=7.98,P <0.01)。褪黑激素还降低了肺和皮肤组织的含水量( F=9.81、 14.79 , q= 3.18~4.66,P <0.05、0.01)。结论 褪黑激素在严重烧伤大鼠中具有很强的抗氧化作用,早期应用能显著降低血管通透性和减轻组织水肿。[关键词] 褪黑激素;烧伤;血管;通透性;水肿;丙二醛;大鼠
