     Effect of Nourishing Yin Antitoxic Capsule 2 on glucocorticoid receptor protein expression in Yin deficiency rats induced by hydrocortisone

    【Abstract】 AIM: To observe the effect of Nourishing Yin Antitoxic Capsule 2 (YC2) on the expression of glucocorticoid receptor proteins (GR) in Yin deficiency rats induced by hydrocortisone, and to investigate how the YC2 relieves the side effects of Glucocorticoid. METHODS: Thirtytwo SD rats were randomly allocated to 4 groups: normal control group, Yin deficiency model control (YD) group, YC2 group and Guifu Dihuang Balls (GFB) group. Radioimmunological method was used to observe the contents of corticosteroid (CORT). Immunohistochemistry was used to observe the expression of GR in the liver cell nucleus. RESULTS: Compared with the normal group, the contents of CORT were higher in YD group (P<0.05), which was (9.92±0.69) μg/L, and the expression of GR in the liver cell nucleus was lower for the optical density ratio of immunohistochemistry stain (0.316±0.017, P<0.05). Compared with the YD group, the contents of CORT in YC2 group were lower(P<0.05), which was (7.28±0.72) μg/L, and the expression of GR in the liver cell nucleus was higher for the optical density ratio of immunohistochemistry stain (0.342±0.013, P<0.05); the contents of CORT in GFB group were little lower(P>0.05), and the expression of GR in the liver cell nucleus was little higher (P>0.05). CONCLUSION: In this experiment, the contents of CORT in YD group were obviously higher than the normal group, and the expression of GR in the liver cell nucleus was lower. The YC2 can increase the expression of GR in the liver cell nucleus, and improve the hyperfunction of hypothalamicpituitaryadrenal (HPA) axis. ......
