VEGF and bFGF induced differentiation of mononuclear cells from human umbilical cord blood into endothelial progenitor cells in vitro

【Abstract】 AIM: To research a way for mononuclear cells from human umbilical cord blood in vitro to be differentiated into endothelial progenitor cells under the induction of VEGF and bFGF. METHODS: The mononuclear cells were obtained from human umbilical cord blood by Ficoll densitygradient centrifugation. The cells were suspended in medium 199(M199) supplemented with VEGF, bFGF for culture, differentiation, proliferation and identification. The expressions of specific antigens on cell surface were analyzed by immunohistochemistry and immunofluorescence. RESULTS: The mononuclear cells from human peripheral blood which were cultured and induced in vitro by VEGF and bFGF were positive for KDR, CD1133 and CD34. CONCLUSION: Mononuclear cells from human umbilical cord blood can be induced to differentiate into endothelial progenitor cells in vitro under certain condition. ......
