反相高效液相色谱法测定连翘中乌索酸和齐墩果酸的含量

     摘要:目的建立同时测定连翘中乌索酸和齐墩果酸含量的测定方法。方法采用高效液相色谱(HPLC)法测定其含量。色谱柱为NovaPak C18(3.9 mm×300 mm,4 μm),流动相为甲醇水(88∶12),流速0.8 ml/min,检测波长210 nm,柱温25℃。结果乌索酸和齐墩果酸分别在0.912~4.560 μg(r=0.999 8)和0.312~1.560 μg(r=0.999 6)范围内峰面积与进样量呈良好的线性关系;乌索酸和齐墩果酸的平均回收率分别为98.2 % (RSD=1.1 %)和97.8 % (RSD=1.4 %)。结论 此法简单、方便、快速、准确,可为中药连翘质量评价和控制提供科学依据。

    Abstract:ObjectiveTo develop a method for the quantitative analysis of ursolic acid and oleanolic acid in Forsythia suspense simultaneously.MethodsHPLC was carried out on the NovaPak C18 column (3.9 mm×300 mm, 4 μm) at 25℃ using MeOHH2O(88∶12) as mobile phase with a flow rate of 0.8 μl and detection wavelength was set at 210 nm .ResultsThere was good linear relationship between the peak area and the sample content injected at the ranges of 0.912~4.560μg(r=0.999 8)and 0.312~1.560 μg(r=0.9996)for ursolic acid and oleanolic acid respectively .The average recovery rate of ursolic acid and oleanolic acid were 98.2 % (RSD 1.1 %)and 97.8 % (RSD 1.4 %),respectively. ConclusionIt's a simple, convenient, quick and accurate method, and can also be used scientifically in the quality estimation and control of Chinese traditional medicine Forsythia Suspense. ......
