高效液相色谱法同时测定苦参片中苦参碱和氧化苦参碱的含量

     摘要:目的建立苦参片中氧化苦参碱和苦参碱的含量测定方法。方法采用高效液相色谱法,使用Thermo色谱柱,以乙腈-磷酸盐缓冲液(取磷酸二氢钾1.36g,加1 ml三乙胺,加水至1000 ml,用磷酸调节pH5.5)(3∶97)为流动相,流速为1.0 ml/min,检测波长为220 nm。结果氧化苦参碱在0.005 5~0.088 mg/ml之间与峰面积积分值呈良好的线性关系(r=0.999 8),平均加样回收率99.41%,RSD为1.28%;苦参碱在0.106~0.170 mg/ml之间与峰面积积分值呈良好的线性关系(r=0.999 9),平均加样回收率99.35%,RSD= 1.25%。结论该法灵敏、准确、专属性强,重现性好,可作为苦参片的质量控制方法。

    Determination of Oxymatrine and Matrine in Kushen Tablets by HPLC

    Abstract:ObjectiveTo establish a method for simultaneous determination of the content of matrine and oxymatrine in Kushen Tablets. MethodsHPLC method was carried out through Thermo C18 column, using acetontrile- phosphate buffer solution (contained 1.36 g potassium dihydrogen phosphate and 1 ml triethylarmine, add water to 1000 ml, adjusted to pH 5.5 with phosphate acid)(3∶97) as mobile phase. The flow rate was 1.0 ml/min with detective wavelength of 220 nm. ResultsThe calibration curve of oxymatrine content was linear within the range of 0.0055~0.088 mg/ml (r=0.999 8),the average recovery of oxymatrine was 99.41%, RSD was 1.28%;the calibration curve of matrine content was linear within the range of 0.170~0.0106 mg/ml (r=0.999 9),the average recovery of Matrine was 99.35%, RSD was 1.25%. ConclusionThe method is simple, fast and accurate, it can increase the quality control index of Kushen Tablets. ......
