摘要:目的利用Burkitt淋巴瘤细胞株Jiyoye细胞探讨转化生长因子β1(TGFβ1)对淋巴瘤细胞生长的抑制作用和cmyc 基因的影响。方法利用MTT及RTPCR方法检测细胞生存率、cmyc 基因的表达水平。结果不同浓度的TGFβ1作用于Jiyoye细胞24 ,48 h和72 h后,细胞生存率随浓度的升高和时间的延长而逐渐降低。实验组各时间段的生存率与对照组相比显著降低(P<0.01),同时,实验组各组间均有显著性差异(P<0.01)。实验组中cmyc 基因的表达24 h开始下降,48 h和72 h时明显受抑制,与对照组相比有显著性差异(P<0.01)。结论 TGFβ1可在浓度和时间依赖性抑制Jiyoye细胞株生长的同时抑制cmyc基因的表达。关键词:转化生长因子β1; Burkitt淋巴瘤; cmyc基因
Effect of Transforming Growth Factor β1 on Cell Growth and cmyc Gene Expression on Lymphoma Cell Line

Abstract:ObjectiveTo investigate the inhibition of transforming growth factorbeta1 (TGFb1) on cell growth and cmyc gene expression on Burkitt lymphoma cell lineJiyoye.MethodsThe cell survival rate and cmyc gene expression were detected by using MTT assay and RTPCR.ResultsAfter 24, 48 and 72 h of different concentration of TGF β1 action on Jiyoye cell line, the cell survival rate decreased along with the increase of concentration and extension of the time. Compared with the control group, the cell survival rate of every stage in experimental group significantly decreased(P<0.01). At the same time, there was also significant difference among the experimental groups(P<0.01). Cmyc gene expression began to reduce at 24 h, and significantly inhibited at 48 h and 72 h ......
