远志蜜炙前后化学成分的对比研究

     摘要:目的比较远志蜜炙前后总皂苷和浸出物的含量;初步比较远志蜜炙前后化学成分的异同。方法以远志皂苷元为对照品,采用香草醛-高氯酸显色法于560 nm波长处分别测定生远志和蜜远志中远志总皂苷的含量;采用药典方法测定和比较远志蜜炙前后70%乙醇浸出物的含量;采用薄层鉴别方法比较生远志和蜜远志正丁醇提取部位化学成分的异同。结果生远志中的远志总皂苷含量为2.096%、蜜远志中的远志总皂苷平均含量为2.207% ;蜜远志的浸出物为50.50%,生远志的浸出物为36.53%。生远志正丁醇部位色谱和蜜远志正丁醇部位色谱基本一致:紫外灯(365 nm)下二者在相应位置呈相同颜色荧光,香草醛硫酸乙醇溶液显色后二者在相应位置的几个斑点均呈紫罗兰色,且远志皂苷元对照品斑点呈紫罗兰色,而在与其相应位置生远志供试品和蜜远志供试品均未检视到相应斑点。结论 远志蜜炙后化痰止咳作用增强的原因在于蜜炙能增加总皂苷的溶出和有效成分的浸出量,蜜炙对远志化学成分的影响在于量变而非质变。

    Comparative Study on Contents of Radix Polygala and the Prepared

    Abstract:ObjectiveTo compare the contents of total onjisaponins and eluate between the crude and the prepared,and compare the chemical constituents between them.MethodsThe contents of total onjisaponins were determined with colorimetry of vanillin-perchloric acid method and detected at 560nm.The 70% ethanol eluate was determined and compared with pharmacopoeia method.The similarities and differences of the n-butanol extractant between the crude and the prepared was compared by TLC.ResultsThe average content of total onjisaponins of the crude was 2.096%, and of the 5 batches prepared was 2.207% ; The average content of eluate of the crude was 36.53%, and of the 5 batches prepared was 50.50%.The TLC of them were concordance.They presented the same color fluorescence in the corresponding position at the ultraviolet light(365nm). After the vanillic aldehyde sulphuric acid ethanol solution developed, violet color spots presented at the same place. Reference substance of Polysaccharide presented violet color ,but in the corresponding position the crude and the prepared didn't show the spots . It proves that the chemical constituents of Polygala Root may not have changed after stir-baked with honey.ConclusionBy stir-baking with honey which can increase the elution of the total onjisaponins and the effective contents may result in the enhance of effect of the Polygala Root, and it can only affect the contents of the onjisaponins but not change the structure of the compounds. ......
