【Abstract】 AIM: To observe the association between the risk of nonsmall cell lung cancers and polymorphism and gene frequency of XRCC 2 C41657T and G4234C in north Chinese Han population. METHODS: The single nucleotide polymorphisms (SNPs) of the C41657T and G4234C in the XRCC 2 gene were genotyped by PCRRFLP analysis in 226 nonsmall cell lung cancer patients and 300 healthy controls. RESULTS: There was XRCC 2 gene C41657T and G4234C polymorphism in north Chinese Han population. The genotype and allelotype distributions of the XRCC 2 C41657T and G4234C SNPs in the nonsmall cell lung cancer patients were not significantly different from those of the controls (all P>0.05). When the two XRCC 2 SNPs were combined, the haplotype distribution in the nonsmall cell lung cancer patients was also not significantly different from that of the controls (P>0.05). But stratification analysis according to histological types and smoking status showed that, compared with C/C genotype, the combination of C/T and T/T genotypes of XRCC 2 C41657T increased the risk of developing nonsmall cell lung cancer (OR: 1.85, 95% CI: 1.02-3.36 ) and that of adenosquamous carcinoma (OR: 2.45, 95% CI=1.05-5.72) in nonsmokers. No association was observed between XRCC2 G4234C and the type of nonsmall cell lung cancers, and also there was no evidence for the interaction between XRCC 2 G4234C and smoking status(P>0.05). CONCLUSION: XRCC 2 gene C41657T allele may be a susceptibility factor for the development of nonsmall cell lung cancer and adenosquamous carcinoma in the nonsomkers of north Chinese Han population. ......
