AR基因启动子区C106T多态性与糖尿病肾病的相关性

     [摘要] ①目的 探讨染色体7q35区醛糖还原酶(AR)基因启动子区C106T多态性与2型糖尿病肾病(DN)相关性。②方法 运用聚合酶链反应限制性片段长度多态性技术(PCRRFLP)、DNA测序技术及高浓度琼脂糖凝胶电泳分离技术,对139例按尿微量清蛋白排泄率(UAER)分为早期DN组(DN+组,61例)和非DN组(DN-组,78例)2型DM病人的AR基因启动子区C106T多态位点的等位基因和基因型进行了检测,并以63例健康成人作为对照(CON组)。③结果 AR基因启动子区C106T多态位点存在T、C两种等位基因和CC、CT、TT三种基因型,DM组T、C等位基因频率和CT、CC基因型频率与CON组差异无显著性(χ2=1.77、1.23,P>0.05);DN+组T等位基因频率和CT基因型频率明显高于DN-组和CON组(χ2=8.63~12.71,P<0.01),C等位基因频率和CC基因型频率明显低于DN-组和CON组(χ2=8.42~14.10,P<0.01)。④结论 AR基因启动子区C106T多态性与糖尿病肾病的发生、发展有关。

    THE CORRELATION BETWEEN POLYMORPHISM OF ALDOSE REDUCTASE GENE AND DIABETIC NEPHROPATHYCHEN WENHUA(Department of Geratology, The Affiliated Hospital of Yanbian University, Yanji 133000, China)

    [ABSTRACT]ObjectiveTo investigate the relationship between polymorphism of C106T at promotor region of aldose reductase(AR) gene in chromosome 7q35 and nephropathy with type 2 diabetes in north China. MethodsA casecontrol study was conducted in 202 Chinese (including 139 cases of type 2 diabetes mellitus with or without nephropathy and 63 normal control subjects). The genotypes and alleles of polymorphism of C106T at promotor region of AR gene were determined by polymerase chain reaction restriction fragment length polymorphism(PCRRFLP) and 4% agarose gel electrophoresis. ResultsThe frequency of the T allele and CT genotype at promotor region of AR gene was significantly higher in DN+ group than that in DN and control(CON) groups (χ2=8.42-14.10,P<0.05),but there was no significant difference between DM and CON groups (χ2=1.77,1.23; P>0.05).ConclusionThe polymorphism of the C106T at promotor region of aldose reductase gene is related to the occurrence and development of diabetic nephropathy. ......
