[摘要] ①目的 探讨染色体7q35区醛糖还原酶(AR)基因启动子区C106T多态性与2型糖尿病肾病(DN)相关性。②方法 运用聚合酶链反应限制性片段长度多态性技术(PCRRFLP)、DNA测序技术及高浓度琼脂糖凝胶电泳分离技术,对139例按尿微量清蛋白排泄率(UAER)分为早期DN组(DN+组,61例)和非DN组(DN-组,78例)2型DM病人的AR基因启动子区C106T多态位点的等位基因和基因型进行了检测,并以63例健康成人作为对照(CON组)。③结果 AR基因启动子区C106T多态位点存在T、C两种等位基因和CC、CT、TT三种基因型,DM组T、C等位基因频率和CT、CC基因型频率与CON组差异无显著性(χ2=1.77、1.23,P>0.05);DN+组T等位基因频率和CT基因型频率明显高于DN-组和CON组(χ2=8.63~12.71,P<0.01),C等位基因频率和CC基因型频率明显低于DN-组和CON组(χ2=8.42~14.10,P<0.01)。④结论 AR基因启动子区C106T多态性与糖尿病肾病的发生、发展有关。[关键词] 糖尿病,非胰岛素依赖型;糖尿病肾病;醛糖还原酶;基因频率
THE CORRELATION BETWEEN POLYMORPHISM OF ALDOSE REDUCTASE GENE AND DIABETIC NEPHROPATHYCHEN WENHUA(Department of Geratology, The Affiliated Hospital of Yanbian University, Yanji 133000, China)
[ABSTRACT]ObjectiveTo investigate the relationship between polymorphism of C106T at promotor region of aldose reductase(AR) gene in chromosome 7q35 and nephropathy with type 2 diabetes in north China. MethodsA casecontrol study was conducted in 202 Chinese (including 139 cases of type 2 diabetes mellitus with or without nephropathy and 63 normal control subjects). The genotypes and alleles of polymorphism of C106T at promotor region of AR gene were determined by polymerase chain reaction restriction fragment length polymorphism(PCRRFLP) and 4% agarose gel electrophoresis. ResultsThe frequency of the T allele and CT genotype at promotor region of AR gene was significantly higher in DN+ group than that in DN and control(CON) groups (χ2=8.42-14.10,P<0.05),but there was no significant difference between DM and CON groups (χ2=1.77,1.23; P>0.05).ConclusionThe polymorphism of the C106T at promotor region of aldose reductase gene is related to the occurrence and development of diabetic nephropathy. ......
