【摘要】 探讨炎性细胞因子与实验性2型糖尿病大鼠及肾脏病变。方法 通过高糖高脂饮食结合小剂量链脲佐菌素的方法建立2型糖尿病大鼠动物模型。4、8周末测量大鼠空腹血糖(FBG)、血清胰岛素(FINs),18周末测量FBG、FINs、转化生长因子β1(TGFβ1)、瘦素(Leptin)、肿瘤坏死因子(TNFα)水平并计算胰岛素抵抗指数(HOMAIR)。用免疫组化、反转录多聚酶链反应(RTPCR)检测TGFβ1在肾脏的表达情况。结果 2型糖尿病大鼠血清TGFβ1、TNFα、Leptin水平明显升高,同时肾脏的TGFβ1表达水平也升高,并伴有肾脏损害。结论 实验性2型糖尿病大鼠存在炎性病变及肾脏损害。【关键词】 2型糖尿病;胰岛素抵抗;炎症;转化生长因子β1;瘦素;肿瘤坏死因子α

【Abstract】 Objective To investigate the changes of inflammatory factors and kidney injury in rat model of type 2 diabetes mellitus Methods The model of type 2 diabetes mellitus was set up by feeding highglucose and highfat diet and by injecting STZ into peritoneal cavity of rat. At the end of 4 weeks and 8 weeks, the fasting blood glucose, the levels of fasting serum insulin were measured. At the end of 18 weeks the fasting blood glucose, the levels of fasting serum insulin, serum TGFβ1,TNFα,Leptin in all the groups were measured. Moreover, immunohistochemistry and reverse transcription polymerase chain reaction(RTPCR) was used to examine the pathological change of kidney and expression of TGFβ1.Results The levels of TGFβ1,TNFα,Leptin in the kidney of the rats of type 2 diabetes mellitus obviously increased. The expressive levels of TGFβ1 in the kidney also increased.Conclusions There are inflammatory injuries in the kidney of rat model of type 2 diabetes mellitus. ......
