立芷雪止血作用量效关系探讨实验研究

     【摘要】 目的 探讨立芷雪发挥止血作用的量效关系。方法 以NIH小鼠和新西兰兔作为实验动物,研究不同剂量的立芷雪对出血时间的影响,同时探讨立芷雪对新西兰兔FDP和FIB的影响。结果当剂量在0.023、0.07、0.21、0.63ku/kg时立芷雪对新西兰兔产生明显的止血作用,但0.63ku/kg组作用弱于0.21ku/kg组,但剂量达到1.90ku/kg、5.71ku/kg 具有明显的抗凝功效;立芷雪在等效剂量时对小鼠出血时间的影响的实验也支持这一实验结果。同时立芷雪0.023、0.07、0.21、0.63ku/kg剂量组对新西兰兔FIB、FDP未见明显影响,但1.90ku/kg、5.71ku/kg剂量组具有明显减少FIB的功效以及明显增加FDP的作用。结论 立芷雪对NIH小鼠产生最佳止血作用的剂量可能在0.58~1.74 ku/kg之间; 立芷雪对新西兰兔产生最佳止血作用的剂量可能在0.21~0.63ku/kg之间,剂量达到1.90ku/kg、5.71ku/kg 具有明显的抗凝作用。

    The dose-response relationship of Reptilase on hemostatic mechanism in rabbits and mice

    【Abstract】 Objective To evaluate dose-response relationship of Reptilase on hemostatic mechanism in rabbits and mice.Methods The effects of different doses of Reptilase on BT, FDP and FIB were examined in animals.Results BT was significantly declined at the dose of 0.023, 0.07, 0.21, 0.63ku/kg for Reptilase, respectively, while the action of 0.21 ku/kg was significantly stronger than that of 0.63 ku/kg; when the dose reached to 1.90 or 5.71 ku/kg, the function of anticoagulation could be exerted significantly; the effect of equivalent dose on BT in NIH mice also supported that in New Zealand rabbit. The effect of Reptilase on FIB and FDP at the dose of 0.023, 0.07, 0.21 and 0.63 ku/kg was not observed in rabbits, the dose of 1.90 and 5.71 ku/kg could reduce the action of FIB and increase that of FDP, respectively.Conclusion The optimal dose of hemostasia action of Reptilase maybe between 0.58 and 1.74 ku/kg in NIH mice and between 0.21 and 0.63 ku/kg in rabbits, and when the dose reached to 1.90 or 5.71 ku/kg, it possess obviously increasing anticoagulation function in rabbits. ......
