    CD51阳性心肌细胞对心肌梗死后小鼠心功能改善的作用

    【摘要】目的探讨CD51阳性(CD51+)的心肌细胞移植治疗AMI后心脏结构及功能变化。方法取C57小鼠14只随机分为对照组(n=7)和细胞移植组(n=7),结扎冠状动脉左前降支制成心肌梗死模型。细胞移植组心肌梗死后立即于心肌梗死部位注射转染红色荧光蛋白的CD51+心肌细胞(RFP-CD51),对照组注射生理盐水。移植后1周及1个月分别用小动物心脏彩色多普勒超声(彩超)仪检测心功能指标的变化及组织形态改变,并且在移植后1个月时观察RFP-CD51细胞是否分化为成熟心肌细胞。结果细胞移植组的小鼠在1周及1个月时的心脏彩超均显示LVEF、左心室缩短分数(LVFS)均显著高于对照组(P0.05)。在心肌梗死3周后发现移植的CD51+细胞分化为成熟心肌细胞。结论AMI后的CD51+心肌细胞移植可分化为成熟心肌细胞、改善心功能、防止心脏重构、减轻心腔扩大。

    Abstract【】ObjectiveTo discuss the cardiac structural and functional changes after CD51+ myocardial cell transplantation in the treatment of acute myocardial infarction (AMI). MethodsFourteen C57 mice were randomly divided into the control (n=7) and cell transplantation groups (n=7). Left anterior descending coronary artery was ligated to establish AMI mouse model. In the cell transplantation group, CD51+ myocardial cells transfected with red fluorescent protein (RFP-CD51) were injected to the myocardial infarction sites immediately after AMI, and phosphate buffer saline was administered in the control group. At 1 week and 1 month after cell transplantation, the changes in the heart function indexes and tissue morphology were detected by small animal ultrasonography. The differentiation of RFP-CD51 cells into mature myocardial cells was observed at 1 month following cell transplantation. ResultsSmall animal ultrasonography revealed that left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVFS) were significantly higher compared with those in the control group at 1 week and 1 month after cell transplantation (both P0.05). The transplanted CD51+ cells were differentiated into mature myocardial cells at 3 weeks after AMI. ConclusionsAfter AMI, transplanted CD51+ myocardial cells can differentiate into mature myocardial cells, which improve heart function, prevent cardiac remodeling and mitigates cardiac chamber enlargement. ......
