miR-223-3p通过靶向NLRP3恢复高血压脑出血大鼠的认知功能

     【摘要】 目的 探索miR-223-3p对高血压脑出血大鼠认知功能恢复的影响及其调控机制。方法 构建高血压脑出血大鼠模型,分析miR-223-3p表达和认知功能改变;过表达/敲降miR-223-3p,探讨其对NOD样受体家族含pyrin结构域3(NLRP3)炎症小体表达和大鼠认知功能的影响;敲降NLRP3,确认该炎症小体对脑出血大鼠认知功能的作用;通过荧光素酶实验鉴定miR-223-3p对NLRP3的直接靶向作用。结果 高血压脑出血大鼠的脑组织中miR-223-3p表达上调。miR-223-3p抑制NLRP3的表达,并恢复大鼠认知功能。荧光素酶实验结果表明miR-223-3p可以直接靶向NLRP3的mRNA并下调其表达。结论 miR-223-3p通过与NLRP3的mRNA结合下调NLRP3的表达,从而恢复脑出血大鼠的认知功能。

    【Abstract】 Objective To explore the effects of miR-223-3p on cognitive function recovery in rats with hypertensive intracerebral hemorrhage (HICH) and its regulatory mechanisms. Methods The rat model of HICH was constructed to analyze changes in miR-223-3p expression and cognitive function.
